About the Stilling

PhD Stilling Investigating Mechanobiology in the Heart

Applications are invited for a 3-year PhD Stilling investigating how cardiac muscle cells (cardiomyocytes) sense mechanical signals, and how these pathways control the cell's structure and function. Based on these insights, the project will seek to understand how mechanical overload in diseases such as heart failure can be therapeutically targeted. The Stilling will be based at the Institute for Experimental Medical Research (IEMR) located at Oslo University Hospital.

IEMR is an innovative and dynamic medical research environment that conducts research focused on understanding the basic mechanisms of cardiac (patho)physiology. With approximately 60 employees and six research groups, our expertise spans molecular biology, physiology and cardiology. The project will be supervised by Professor William Louch.

Work tasks

The PhD Stilling will be 3 years in duration and, depending on starting date, have latest completion of the end of 2029. Funding will be provided by a grant from the South-Eastern Norge Regional Health Authority.

The heart is highly mechanosensitive, allowing it to "remodel" when its workload changes. The Louch group has shown that workload is critically sensed by cardiomyocytes. With a moderate increase in workload, such as during exercise, compensatory cardiomyocyte remodeling occurs and cardiac function improves. However, there is a "tipping point", beyond which high workload triggers pathological remodeling, leading to impaired cardiac performance and heart failure. It is unknown how such changes are triggered, and how these signals can be therapeutically targeted.

The current project aims to address these issues using living myocardial slices; a system which allows the culture of human tissue to investigate the tissue response to predefined workloads. It is hypothesized that the tipping point represents a switch, where a novel set of sensory proteins are activated to trigger pathological remodeling. Using these data, the project will identify and test the efficacy of novel mechanosensitive targets, using tissue from heart failure patients. Thus, the project will provide proof-of-principle for a new class of therapies aimed at treating heart failure by directly targeting the mechanical signals underlying this disease.

Qualifications

  • Applicants must hold a BSc in Biology, Biomedical Sciences, Engineering, or Physics
  • Previous experience with:
  • Advanced light microscopy techniques including confocal imaging
  • Qualitative and quantitative analysis of microscopy images, using tools such as ImageJ
  • Experimental testing of genetic alterations
  • Fluent oral and written communication skills in English are required
